Description

Piroctone olamine (trade name Octopirox) is a second-generation hydroxypyridone antifungal agent widely used in anti-dandruff shampoos, scalp treatments, and dermatological formulations targeting Malassezia-associated conditions. It was developed as a successor to ciclopirox olamine, offering improved efficacy and tolerability at lower concentrations. Piroctone olamine shows potent fungistatic and fungicidal activity against Malassezia species (M. globosa, M. restricta, M. furfur), the lipophilic yeasts implicated in seborrheic dermatitis, dandruff, and Malassezia (pityrosporum) folliculitis. It is considered one of the most effective cosmetic-grade antifungals available, permitted at up to 1% in leave-on and rinse-off formulations in most regulatory regions.

Mechanism of Action

Piroctone olamine acts primarily by chelating iron (Fe³⁺) and other polyvalent metal ions essential for fungal mitochondrial electron transport chain function. By sequestering iron from cytochrome b and other iron-sulfur proteins in the mitochondrial respiratory chain, it disrupts oxidative phosphorylation and energy production in fungal cells, leading to cell death. Additionally, piroctone olamine inhibits fungal cell membrane ergosterol biosynthesis and disrupts membrane integrity. It also inhibits the formation of fungal biofilms on the skin surface. Against Malassezia species, it reduces the metabolism of sebum triglycerides into irritating unsaturated fatty acids (notably oleic acid), thereby addressing a key pathogenic trigger in seborrheic dermatitis. Its sebostatic properties further reduce the lipid-rich environment that supports Malassezia colonization.

Side Effects

  • Known hypersensitivity to piroctone olamine or hydroxypyridone derivatives
  • Avoid contact with eyes (irritant to mucous membranes)
